The REALQUALITY RQ-HIV DNA is a device indicated for the detection and quantification of the total DNA of the human immunodeficiency virus (HIV-1), group M, by Real-Time PCR.

The human immunodeficiency virus (HIV-1) establishes, from the early stages of infection, a provirus reservoir formed by viral genomes integrated into the host cells, partly defective and partly able to initiate new replicative cycles. This reservoir mainly resides in T CD4 + memory lymphocytes and rapidly fuels the restart of viral replication at the end of the interruption of antiretroviral therapy (ART).
Although the different forms of DNA (total, integrated and 1 or 2-LTR) may represent different entities in the viral replicative cycle, there is a strong positive association between total and integrated HIV-1 DNA, especially in patients with chronic infection. This supports the observation that the integrated form is the most widespread during ART and that total HIV DNA is therefore a representative biomarker of the HIV-1 reservoir.
The total amount of HIV DNA in the blood or in peripheral blood mononuclear cells (PBMC) provides information on both the pathogenesis and the course of infection. Both the defective viral and replication genomes participate in the activation of the host cells and the progression of the disease, thus influencing the course of the infection itself. In untreated patients, total HIV DNA is an indication of progression to the full-blown disease independent the viremia (HIV RNA) and the number of CD4 cells.
